For proper serodiagnosis of bovine tuberculosis, knowledge regarding dynamic changes in humoral immune response throughout the disease spectrum is quite needed. With the aim of studying kinetics of seroreactivity of Mycobacterium bovis AN5 in experimentally inoculated cattle, sex and age matched 4 healthy cattle were inoculated with live M. bovis AN5 cells, keeping two animals as control. To know the kinetics of antibody level/ titre against M. bovis AN5 culture filtrate antigens, screening of all experimental sera was performed by ELISA starting from 0 day through 45th week post inoculation (P.I.) It was observed that most of the sensitized cattle showed elevated serum antibody titre 20th week P.I. onward. To assess the seroreactivity of the sensitized cattle at the polypeptide level, Western blot analysis was employed using the sera of sensitized cattle in different stages post sensitization, viz. 0 day, 20th week and 45th week P.I. The immunodominant polypeptides of molecular weights 95 and 65 kDa were found more common in seroreactivity at 20th week P.I., whereas that of molecular weights 70, 65, 52, 38 and 25 kDa were observed much similar in seroreactivity at 45th week P.I. among sensitized animals. Further study is needed to explore the potentiality of the immunodominant polypeptides reported while preparing serodiagnostic tool(s) against bovine tuberculosis.
