Isolation and Characterization of Cellulolytic Bacteria from a Biofertilizer Generated from Municipal Solid Waste

The study analyses a biofertilizer generated from Municipal Solid Waste (MSW) for the presence of cellulase producing bacteria. The biofertilizer was serially diluted and enriched to get a bacterial count of 214 x 10^ CFU/mL. Eighteen colonies were selected for flirther study based on morphological characterization and pure cultures were made out of it. The isolated colonies were subjected to an initial screening for cellulase producing bacteria in CMC agar plates. Out of the eighteen colonies isolated only one of the isolate was found to produce a clear zone in plate assay on flooding with Congo red (1% w/v). Crude extract was taken from the supernatant of the broth culture of this organism after centrifugation and enzyme assay was done. The crudeenzyme was further optimized by varying the incubation time, temperature of incubation and pH of the medium. The results show that the crude enzyme shows maximum activity at 40°C, at neutral pH and with an incubation time of 48 hours.
