Based on carbohydrate recognition, three lectins were isolated from Olneya tesota (Palo fierro) seed extracts using a tandem system of two affinity chromatography columns (galactose and immobilized fetuin). PF1 lectin has 45 kDa subunits while PF2 and PF3 have subunits of 33 and 66 kDa respectively. Under native conditions these lectins are glycoproteins that migrate as dimers (PF1), tetramers (PF2) and monomers (PF3). As most lectins from cultivated legumes, PF1 presented affinity for free monosaccharides. On the contrary, PF2 and PF3 agglutinins were specifically inhibited by complex carbohydrates such as those present in fetuin, mucin and immunoglobulins. Analysis of amino-terminal sequences showed 60 to 80% homology of PF2 to α amylase inhibitors and to PHA (Phaseolus vulgaris lectin), while PF3 sequence did not resemble to any published lectin.
