In vitro plant regeneration studies in three indica rice varieties

Genotypic effects were observed for embryogenic callus induction and subsequent plant regeneration in three Indica rice varieties. Plant regeneration was investigated from seed derived callus cultures in three rice varieties viz. PR118, PR114 and PR120. Seeds were cultured on Murashige and Skoog (MS) medium supplemented with 2, 4-D (4.0 mg/1) + BAP (0.2 mg/1). PR 118 exhibited a high embryogenic callus induction efficiency (81.34) followed by PR 114 (50%) and PR 120 (33.21%). Calli were maintained for 45 days on the same medium by regular subculturing after 15 days. Then they were transferred to the shoot regeneration medium with various concentrations of NAA (Napthelene acetic acid) and BAP (Benzyl amino purine). Plant regeneration frequency was found the highest (78.0%) on the PR 118, when NAA, BAP with gelrite were used @ 0.5 mg/1, 4.0 mg/1 and 0.2% respectively. Half strength MS basal medium was found best for root regeneration from in vitro raised shoots. The plantlets were able to regeneratewithin 8-10 weeks.
