摘要

Isolation of Mjctbatterium paratubercitltsis, the ctiological agent of Johne's disease on artificial medium is difficult. Previously isolated 10 isolates of M. paratuberculosis of goat and 4 of sheep origin were passaged and maintain-d on Herrold's egg yolk medium with mycobactin ). These 10 isolates were attempted to adopt and grow in modified Watson & Reid liquid medium. Present study undertaken to standardize the air requirement of these isolates, three air columns viz- 25% (75% medium 4- 25% air), 50% (50% medium + 50% air) and 75% (25% medium + 75% air) were used with two different pH of medium i.e. 5.8 and 7.4. After an interval of 4 weeks one isolate from sheep and one from goat were found to grow successfully in liquid medium with 75% air column and pH 5.8. The above observitions indicate that to grow M. purituberculosb on solid medium and maiatain in subsequent passages on the Herrold's egg yolk medium is easy but its growth into the liquid medium poses a problem and requires further investigation.
