Acrosome reaction test was used as a tool to determine in vitro fertilizing ability of unwashed and washed bubaline spermatozoa frozen in EYT dilutor fortified with 0, 5, 10 and 15 mM of caffeine (EYT-C_0, EYT-C_1, EYT-C_2 and EYT-C_3) for comparing performance of the dilutors to determine the optimum concentration of caffeine for cryopreservation of buffalo spermatozoa. Acrosome reaction percentage was significantly affected by washing, bulls, freezing stages, dilutors and interactions of washing xbulls washing x freezing stages and washing x bulls x freezing stages. Motility, livability and sperm abnormality were also measured simultaneously to evaluate the same samples. At post-thaw stage, acrosome reaction was significantly (P<0.05) higher in EYT-C_1 for both unwashed and washed samples as compared to control which indicated that caffeine addition @ 5 mM is significantly beneficial for cryopreservation to get the highest fertilizable spermatozoa. Prgressive sperm motility and livability were also highest in EYT-C_1. The sperm abnormalities were within permissible range in all dilutors at all stages of freezing.
