Abstract.The haemagglutination (HA) and haemagglutination‐inhibition (HAI) techniques using CrCl3to couple purified HBAg to human red cells were standardized and modified as follows:1Selection of human donor red cells used for coupling purified HBAg with CrCl3. It was found that the red cells of different human donors vary greatly with regard to their ability for optimal coating with HBAg.2Improvement of the CrCl3coupling technique by which much less purified HBAg is used.3The use of a preservation fluid, developed in the Central Laboratory of the Netherlands Red Cross Blood Transfusion Service, in which the HBAg‐coated cells can be preserved up to 4–6 weeks without change of titre in HA and HAI when tested against standard sera.By these modifications, purified HBAg can be reproducibly coupled to human red
