THE STRATEGY USED to increase throughput in the laboratory at Pfizer Central Research (Sandwich, Kent, U.K.) involved the development of an LC-MS-MS screening technique to monitor the transport of multiple compounds across Caco-2 cell monolayers. The evaluation of multiple compounds in parallel, enabled by the selectivity of MS-MS, was found to reduce the interexperiment variability associated with earlier single-compound methods. However, in a number of multiple-compound screens it was noted that some compounds could apparently enhance the absorption of others, giving eroneous results. Positive results indicated in rapid multiple-compound screens therefore require subsequent verification in single-compoond experiments.
