摘要

An amide hydrogen in polypeptide main chain exchanges with bulk solvent hydrogen at various rates depending on its physical and chemical environment. When an amide hydrogen is fully exposed to the bulk solvent, it typically exchanges very rapidly (milliseconds to seconds at neutral pH at room temperature). It exchanges much more slowly when the amide hydrogen is not accessible from bulk solvent by hydrogen bonding, etc. When the polypeptide is dissolved in deuterated water, the main chain amide hydrogen will gradually exchange with bulk deuterium. This amide hydrogen/ deuterium-exchange (H/D-Ex) phenomenon has been used to study protein dynamics for several decades.
