High-Sensitivity Detection of Prostate Specific Antigen

The advantages of flexible and easy protein detection through enzyme-linked immunosorbent assay (ELISA) are combined with the sensitivity of the exponential signal amplification typical for PCR in Imperacer~(tm) technology (Chimera Biotec GmbH, Dortmund, Germany). In this technology, antibody- DNA conjugates were used as a substitute for the antibody - enzyme conjugates generally utilized in an ELISA.~1 By using the amplification of the marker DNA, typically a 100-10,000-fold increase in ELISA sensitivity was observed.~(2,3) The technology comprises a set of antibody-DNA conjugates for maximum binding efficiency, and is available as species-specific secondary conjugates (e.g., anti-rabbit, -goat, and -human),~4 as well as customized primary conjugates for selected targets.~5 The technology can be used for human and veterinary diagnostics, biopharmaceutical research, and food and beverage control, and finds application in, for example, the detection of viruses and neurodegenerative diseases, cancerdetection and monitoring, quality assurance, and pharmacokinetics.
