As morphological skills are decreasing, new options are needed for the detection and identification of PCN species. At the same time, needs for analysis are increasing because of new regulations at the European Union scale. In this study, one option based on molecular detection of PCN cysts was evaluated. Three molecular tests, either species specific PCR or PCR-RFLP were evaluated for their sensitivity and specificity while testing several cysts at the same time. Samples composed of five and even 10cysts, of either one or a mixture of both PCN species, can be correctly detected and identified. Complementary procedures, such as simplex use for duplex species, specific PCR or dilutions of DNA extracts, could really improve the detection process.
