Chicken erythrocytes, labelled with 51Cr and coated with thyroglobulin, were pretreated with heat inactivated serum from guinea pigs immunized with thyroglobulin in complete Freund’s adjuvant. When incubated with purified spleen or peripheral blood lymphocytes from normal guinea pigs, the red cells were lysed, as assessed by isotope release. Presence of non-lymphocytic cells enhanced the initial rate of the reaction. The cytotoxic reaction was inhibited by pretreatment of the effector cells with antimycin A or with concanavalin A, respectively. The activity of the sera was mediated by a γ2antibo
