IL-2, a glycoprotein (143 aa), is potent immune system modulator affecting nearly every facet of host immune response. Full length mRNA of broiler parent stock of Indian origin was cloned and sequenced. Total RNA from induced PBMCs incubated for 2 h was isolated and cDNA was synthesized by RT-PCR. Using specific primers complete mRNA was amplified and cloned in pGEMT. The nucleotide sequences were analysed using different softwares. Complete ORF was comprised of 432 sequences. SDL-IL2 showed high genetic identity (98.6 to 99.8%) with other chicken stocks. Muscovy duck revealed highest distance from SDL (29.4%). SDL showed least similarity (26.2%) with mouse and highest similarity (31.5%) with cattle. A total of 532 nucleotides from 19 species were aligned. In SDL broilers the transversions as compared to consensus sequences was observed at 51nt (T~R C) positions. SDL showed differences in nucleotide bases as compared to other chicken sequences at 51, 9, 95, 101, 152 and 421nt positions. Muscovy duck showed highest nucleotide differences (160 out of 485) as compared to consensus sequences. The phylogenetic tree revealed that all the avian species made one cluster and all the mammalian species clustered separately making another cluster.
