摘要

The ultrastructural changes produced by Mesoridazine and Nortriptyline in neurons and gliain vitroare reported. Concentrically laminated bodies (CLB) and increased numbers of dense bodies were produced by exposure to these drugs. The number and complexity of the CLB increased with increased dose and longer timein vitroand their formation appeared to be reversible.Numerous investigators have reported increased numbers of dense bodies and concentrically laminated bodies (CLB) in tissuesin vivoandin vitrotreated with certain drugs. Neurons and gliain vitrotreated with chlorpromazine, (2) LSD, (6,13) chloroquine (4) and bilirubin (15) develop these lipid inclusions. Neurons and muscles of rabbits and rats treated with chloroquine, (8) dorsal root ganglia and retinal neurons of animals treated with tricyclic antidepressants and chlorphentermine, and lymphoid tissue of rats treated with chlorphentermine also developed CLB. (9,10,11) CLB have also been found in the substantia nigra of newborn Gunn rats, (1) neurons and glia of patients with Tay-Sachs disease, (16) and other diseases. (7)We have studied the dose and time relationships of the development of CLB and dense bodies in neurons and gliain vitrowhen treated with the tricyclic antidepressant nortriptyline and the phenothiazine mesoridazine.
