The biochemical changes in wheat (Triticum aestivum) plant induced by NaCl stress was investigated by evaluating the activity of alpha -amylase and content of soluble sugar, proline, chlorophyll and carotenoid along with protein banding pattern of leaf tissues in relation to amelioration of salt stress by seed soaking treatments. Three level of NaCl and four seed soaking treatments i.e. Distilled water (DW), Salicylic acid (SA-1 mM), Sodium thiosulphate (ST-1 mM) and Thiourea (TU-5 mM) were used for 15 days after germination. Control plants received natural soil EC 1.4 dSm-1. High salt level (10 dSm-1) caused rapid reduction in plant growth. Seed soaking treatments caused a significant recovery in plant growth rate as well as decreased the activity of salt induced alpha -amylase and also the content of soluble sugar, proline, chlorophyll and carotenoids. SDS-PAGE analysis of leaf protein extract showed salt and chemicals induced de novo expression (35, 40, 70, 75, 85 kD) and repression (25 KD) of proteins. These results suggest that seed soaking treatments effectively alter the salt induced biochemical component in wheat plants and help in recovery of plants to overcome moderate salt stress effects.
