Molecular detection and characterization of Nucleocapsid protein gene of Groundnut Bud Necrosis Virus (GBNV) Isolate from Tomato
Plant showing necrotic rings with green hallow on leaves of tomato were collected from the field and mechanical inoculated to Vigna unguiculata cv. C-152. plants which started expressing chlorotic lesions, three to four days after inoculation. RT-PCRwas performed after total RNA extraction from infected chilli leaves. A fragment of approximately 831bp corresponding to the NP gene was amplified, cloned and sequenced. The nucleotide and amino acid sequence of NP gene of GBNV-Raichur isolate shared highest nucleotide identity of 93.10 to 98.90 and amino acid identity of 94.90 to 99.60 with the other GBNV reported isolates. The result of phylogenetic analysis of nucleotide and deduced amino acid sequences of current isolate under study with other tospo viruses clearly indicated that, PBNV- Raichur is closely clustered with PBNV and GBNV isolates reported earlier with respect to both nucleotide as well as amino acid analysis.
