摘要

In this study,the effects of 2,4-Dichlorophenoxyacetic acid (2,4-D),one of the most used growth regulators in plant tissue culture,on oat embryo culture was investigated.Mature embryos of 2 oat cultivars (Faikbey and Seydisehir) were used as explant sources.The effect of,five different doses of 2,4-D (0,2,4,6 and 8 mg l-1) on callus induction,callus weight,regeneration capacity and culture efficiency parameters were examined in vitro conditions.In the parameters examined for both cultivars,the application of 2 mg l-1 2,4-D gave the highest results.As a result of cytological studies with root samples obtained from regenerants,it was determined that the dose of 2 mg l-1 2,4-D did not cause any change in the number and structure of chromosomes.In oat mature embryo culture studies,it has been determined that a dose of 2 mg l-1 2,4-D provides the highest tissue culture parameters and can be used safely as it does not cause any somaclonal variation.
