The effect of cryopreservation using a Tris-based extender supplemented with different concentrations of reduced glutathione (GSH), catalase (CAT), alpha-lipoic acid (LALA) and superoxide dismutase (SOD) as antioxidants on the post-thaw freezability, mitochondrial function, DNA integrity and freezing potential of buffalo semen was determined. It was shown that cryopreservation significantly decreased the evaluated parameters, while the addition of antioxidants had a beneficial effect in a dose-dependent manner. Addition of 10 mM GSH, 100 U/ml CAT, 15 mM ALA or 50 U/ml SOD significantly increased post-thawing sperm motility and viability, maintained acrosome integrity and improved mitochondrial function and DNA integrity. Addition of 10 mM GSH, 100 U/ml CAT or 50 U/ml SOD increased the In vitro embryo development to the blastocyst stage of oocytes fertilized with the extended sperm and augmented the natural pregnancy rate. In conclusion, the addition of antioxidants improves the characters of extended buffalo semen.
