Surface plasmon resonance (SPR) spectroscopy has been used for detection of DNA hybridization. This method allows label-free detection and in-situ detection. However the detection sensitivity is low for DNA hybridization. Our previous work demonstrated that the colloidal gold attached probe DNA monolayers could enhance the SPR signal when DNA monolayes is surrounded by air. In this work, we examined the SPR measurements of hybridization adsorption at the particle attached DNA monolayer system in aqueous solution by SPR measurement. In the presence of colloidal gold, the SPR signal was clearly enhanced compared to that found in non-attached probe DNA monolayer.
