Conversion of cytoplasm type in sugarbeet (Beta vulgaris L) during inbreeding. I. Hybrid analysis and testing for mitochondrial DNA type.
Male sterility (MS) was studied in 8 families of sugarbeet lines obtained by inbreeding from a single, self pollinated plant. Inbreeding coefficients in these families varied from 0.5 to 0.916, and of 401 plants studied, 68 had sterile pollen. The frequency of MS plants increased from 0.029 to 0.4 after inbreeding. Genetic analysis showed that MS was determined by cytoplasmic inheritance. Analysis of mtDNA digested with BamHI and EcoRI, and blots of digested mtDNA probed with COXII, COB and Ndl, revealed that N- and S-cytoplasm types contained corresponding mtDNA types. The possibility of converting N-cytoplasm into S-cytoplasm by means of a nuclear mutator gene is discussed.
