Phylogenetic relationships among cultivars, species, and hybrids in the genus Fagopyrum Mill. assessed by RAPD analysis.
Twenty-two buckwheat genotypes, including seventeen cultivars/lines of Fagopyrum esculentum, two F. tataricum lines, one F. cymosum line, one F. giganteum line (F. tataricumXF. cymosum hybrid), and one hybrid clone between F. esculentum and F. cymosum, were studied by RAPD analysis. Eleven primers generated 122 DNA fragments, 114 of which were polymorphic. RAPD markers specific to some F. esculentum cultivars were found. In a dendrogram constructed using indices of dissimilarity, F. esculentum cultivars formed two main clusters in accordance with their origin. Four cultivars selected at the Hokkaido Experimental Station (Japan) comprised a separate cluster. Interspecific polymorphism was considerably higher than differentiation between F. esculentum cultivars. F. esculentum and F. tataricum were most divergent genetically; F. cymosum was closer to F. esculentum than to F. tataricum. Interspecific hybrids possessed fragments specific to both parental species, which proved their hybrid origin at the molecular level.
