Effect of non-steroidal anti-inflammatory drugs on bone marrowstem cells in male Albino mice.

NSAIDS may generate free radicals which coulddamage genetic material. Groups of 10 mice each were injected intraperitoneal nonsteroidal antiinflammamtory (NSAIDS) alone or in combinations, once and sacrificed 24hlater for evaluation of bone marrow chromosomal aberrations (CA), and micronuclei(MN). We examined 5000 stem cells from each group of 10 mice. Control cells had 2.2-2.4% MN and 13-13.8% CA. Results showed that mice given 30mg Diclofenac, 3mgMeloxicam, 5mg Ketoprofen, 30mg Ibuprofen or 0.3mg/kg Piroxicam had increased CA(~20-32%) and MN (3.8-4.5%) whereas in those injected 30mg/kg Naproxen we found nosignificant increase. However, when combined with 30mg iboprufen, 30mg/kg naproxendid increase CA and MN. Combinations of 5mg Ketoprofen with 0.3mg/kg Piroxicam0.3mg/kg or Meloxicam 3mg/kg + Diclofenac 30mg/kg enhanced MN (6.0-6.3%) and CA(41-41.6%) compared with single NSAIDS or controls. This may have been an additiveand/or synergistic effect.
