Genetic variation in a Caspian horse population determined by AFLP Markers.

In order to assess genetic variation within the Caspian horse breed in Iran, we performed an amplified fragment length polymorphism (AFLP) analysis. Thirty two individuals were analysed using five AFLP primer combinations which produced 52 polymorphicbands. Average NeiJs Genetic heterozygosity, average Uniformity Index of the AFLP fingerprinting pattern within a population and Polymorphism information content (PIC) per marker were determined (calculated assuming the population was at Hardy-Weinbergequilibrium). The highest and the lowest PIC values occurred with EXT2 (0.3224) and (0.2846), respectively. The values of average heterozygosity in our study was 0.367 and highest unbiased expected heterozygosity was found in E2T2 (0.4144) while lowest was detected in E1T1 (0.3506). The average number of effective alleles (ne) in primer combination was 1.6. Genetic similarity was calculated according to Jaccard similarity Index and Simple Matching coefficient. Individual dendrogram constructed using the UPGMA analysis clearly distinguished the 32 individuals. The findings suggest that AFLP analysis revealed genetic relationships and variation amongst Caspian horses.
