Effect of calciferol, calcium and metformin on type 2 diabetes mellitus in male rabbits.

Type 2 Diabetes mellitus was induced in rabbits with 120mg/kg alloxan and 50mg/kg nicotinamide injected intraperitoneally (IP). The condition was confirmed by fasting blood glucose (FBS) of ≥ 200 mg/dl and insulin resistance >3mg/dL at 2 weeks. We then intubated groups of 5 rabbits each with 100IU Vitamin D (VitD) with or without 500mg/kg calcium gluconate (Ca) or 11mg/kg metformin daily for 30 days. Controls consisted of 5 diabetic and 5 normal rabbits given distilled water. We assessed clinical status and body weights, serum FBS, insulin, insulin resistance, interlukein-6, calcium and VitD levels before or 14 and 30 days after induction of diabetes. Compared with controls (P < 0.05), by 30 days body weights declined 28-30% in diabetic rabbits but less in those given VitD (-11%). By 14 days FBS increased ~56% in diabetic rabbits, but by 30 days all treated rabbits had normal FBS. Serum insulin declined 15-38% in all groups except for those given VitD. At 14 days, we found large rises (~40%) in insulin resistance, but by day 30, rabbits given VitD had normal values. Similar results were found with Interleukin-6. At day 14 there were large declines in serum Ca -68% in treated or not diabetic rabbits but by 30 days not in those given VitD. Our findings suggest that Vitamin D supplementation ameliorates effects of type 2 diabetes mellitus in rabbits.
