A simple purification method of bean common mosaic virus.

Bean common mosaic potyvirus (BCMV) was purified from Phaseolus vulgaris in Egypt by a quick and simple method which preserved infectivity. Aggregation and fragmentation of the virus particles was prevented by using n-butanol for clarification followed by polyethylene glycole (PEG 6000) precipitation then pelleting the virus by high speed centrifugation. This method was quite efficient for the purpose of studying the morphology of the virus particles and some chemical analysis. This technique yielded large quantities of virus particles free from any contamination. Electron micrographs showed that the viruses were flexuous filamentous particles of 840 nm long and 13-15 nm wide, of fine striation. A central channel could not be seen and the basic helix was also obscure. The viral capsid was shown by SDS PAGE to consist of 1 protein band and the molecular weight of the protein subunit was 34 kDa.
