Occurrence of peach latent mosaic viroid in apple (Malus domestica).

A small RNA with the structural and functional properties characteristic of viroids was isolated from apple fruit cv. Golden delicious and pear bark and fruit. It induced canker on bark, scab and streak disease of fruit. Viroid detection in nucleicacid extracts of these tissues was confirmed by reverse transcription polymerase chain reaction (RT-PCR) followed by southern and dot blot hybridization analysis of the amplified products with peach latent mosaic viroid PLMVd cRNA probe. Analysis by southern and dot blot hybridization suggested that the disease agent shares similarities in sequence with peach latent mosaic viroid (PLMVd) and no similarities with apple scar skin viroid (ASSVd). These products were absent from amplified extracts of uninfected tissues ones. PLMVd infected apple plants have been sporadically detected in commercial orchards of apple and pear.
