Double trouble for Langerhans cell histiocytosis

In this issue of Blood, Sengal et al provide a detailed phenotypic and functional analysis of the inflammatory infiltrate in the Langerhans cell histiocytosis (LCH) lesion. Using a BRAFV600ECD11C preclinical model of systemic LCH, they demonstrate the synergistic effect of dual treatment with a MAPK inhibitor directed toward the pathologic LCH cell and checkpoint blockade with an anti-programmed death-1 (alpha-PD-1) antibody directed toward the exhausted T cells in the LCH lesion microenvironment (see figure).(1)
