摘要

Direct shoots regeneration in high frequency was induced in hypocotyl explants of Ailanthus excelsa. About 10-15 shoots were regenerated from a single explant cultured on full strength MS medium containing 1.61 mu M NAA + 4.44 mu M BAP and supplemented with ascorbic acid 100 mg/l and citric acid 50 mg/l. The shoots were further multiplied on half strength MS medium augmented with 0.57 mu M IAA + 17.7 mu M BA. For elongation of shoots these were transferred on to medium containing 0.57 mu M IAA + 8.88mu M BA. Maximum rooting (50%) was observed when excised shoots were transferred to % strength MS medium containing 5.37 mu M NAA and 2.32 mu M Kn for rooting. The plantlets were acclimatized nnder in-vitro conditions and subsequently shifted to in poly-mist house and maintained there for about a month. These plants were kept in shade house for another month prior to their transfer in the fiejd. The protocol developed is not only suitable for rapid multiplication but also for physiological studies associated with adventitious shoot regeneration.
