Effect of induction media on callus formation in un-pollinated ovule culture of three melon cultivars

Production of inbred lines, such as double haploid plants which are produced from pollen or ovule culture, is an important tool in a crop breeding program. Some factors affecting in vitro gynogenesis, including genotypes of donor plants and inductionmedia on callus formation in the un-pollinated ovule culture of melon, were investigated. A 3x3 factorial experiment in completely randomized design with 5 replications, 2 factors including 3 melon cultivars in Thailand ('Green Net', 'Honeydew" and 'PotOrange") which were used as donor plants, and 3 induction media [Ml, MS + 0.20 mg L1 6-benzylaminopurine (BAP) and 0.40 mg L1 1-naphthalene acetic acid (NAA); M2, MS + 0.30 mg L1 BAP + 0.30 mg L1NAA; and M3, MS + 0.40 mg L1 BAP + 0.60 mg L1 NAA] was conducted. Un-pollinated ovules from melon flowers at one day before flowering were used and transferred to induction media in dark condition for 4 days, then their explants were cultured on daily 16 h LED light and 8 h dark, 80 umol nv2 s1. Callus formationwas measured after 6 weeks. The interactions between the 2 factors were non-significant. The callus formation potential of all the 3 melon cultivars showed to be significant. The addition of 0.20 mg L1 BAP and 0.40 mg L1 NAA into the MS medium induced the highest percentage of explants with callus formation with an average of 75.54%. Callus formation percentages, achieved in melon cultivars, can be indicator of efficient production of doubled haploids of melon in the future.
