Plant regeneration from a tissue culture system is often the most critical step in the success of various biotechnological techniques. An efficient adventitious shoot regeneration protocol for 'Elita' plum rootstock ([Prunus pumila L. x p. salicina Lindl.) x [P. cerasifera Ehrh.)) was established. The method was based on an auxin pretreatment of in vitro leaf explants applied before placement on a regeneration medium under standard culture conditions. A high percentage of explants (up to 68.9%) showing shoot regeneration was obtained from leaves pre-cultured for 4 h in liquid MS medium supplemented with 2.5-5.0 mg L1 indole-3-acetic acid. The protocol presented here could facilitate the application of genetic engineering methods to produce mature plum trees with improved resistance to biotic and abiotic stresses.
