Ultrasound-assisted extraction was used to extract crude polysaccharides (PS) from Cordyceps sinensis (Cs-4) mycelium, and the effects of processing parameters on polysaccharide yields were optimized by random-centroid optimization (RCO). The optimal extraction conditions were as follows: liquid to solid ratio of 30 mL/g, ultrasonic power of 550 W ultrasonic time of 20 min, and extraction time of 3. Under these conditions, the maximum extraction yield of PS was 1660%. The partially purified PS-1 contained high-molecular weight (730 kDa, 80.86%) and low-molecular-weight (5.8 kDa, 19.14%) polysaccharides composed of D-glucose, D-mannose, and D-galactose in a ratio of 1.8:1.2:1.0 with cc-type glycosidic bonds. The results indicated that ultrasound-assisted extraction was a very powerful and efficient tool for the extraction of polysaccharides from fungi or mushrooms, and RCO was an useful approach for experimental variable optimization.
