Expression, Correlation, and Clinical Significance of DACH1 and ER alpha in Human Endometriosis Tissue

OBJECTIVE: To study the expression, correlation, and clinical significance of DACH1 and ER alpha in endometriosis tissue. STUDY DESIGN: Reverse transcription polymerase chain reaction (RT-PCR), real-time PCR, and immunohistochemistry were used to assess DACH1 aand ER alpha mRNA expression levels in 51 cases of ovarian endometriosis, 11 cases of adenomyosis, 9 cases of ectopic peritoneal tissue, 17 cases of eutopic endometrium, and 30 cases of normal endometrium. A follow-up study of all patients' relapse rates was conducted. RESULTS: We found that the expression rates of DACH1 in ovarian ectopic endometrium (34/51, 66.7%), adenomyosis (6/11), and ectopic peritoneal tissue (6/9) were significantly lower than in normal endometrium (28/30, 93.3%) and eutopic endometrium (14/17; p0.05). The rate of ER alpha expression in ovarian ectopic endometrium (32/51, 62.7%), adenomyosis (5/11), and peritoneal ectopic tissue (5/9) were significantly lower than in normal endometrium (27/30, 90%). Furthermore, the expression rates of DACH1 and ER alpha mRNA in endometriosis were significantly lower than in the endometrial tissue (p 0.05). The data demonstrated that the expression of DACH1 and ER alpha are negatively correlated. Finally, we found that rate of recurrence for DACH1-positive patients was lower than that for DACH1-negative patients. CONCLUSION: We confirm the findings that DACH1 is less frequently expressed in endometriosis than in normal endometrium and suppresses the function of ER alpha. These findings are important for the study and diagnosis of endometriosis occurrence and recurrence.
