Hypocotyl explants cultured on M.S. medium supplemented with different IAA (1.0 mg/1), NAA (1.0 mg/1), 2, 4-D (1.0 mg/1) alone produced white callus. Whereas callusing was more pronounced in 1.0 mg/1 IAA, compared to NAA (1.0 mg/1) and 2,4-D (1.0 mg/1). Hypocotyl explants cultured on M.S. medium supplemented with BAP (1.0 mg/1) or Kn (1.0 mg/1) alone showed better response in shoot differentiation with green callus. However, BAP (2.0 mg/1) showed good results compared to Kinetin (2.0 mg/1) in inducing shoot buds. IAA (1.0 mg/1) + BAP (2.0 mg/1) and IAA (1.0 mg/1) + Kn (2.0 mg/1) gave good response and produced shoots along with green nodular callus with more number of shoots buds. The combination of IAA (1.0 mg/1) .+ BAP (5.0 mg/1) found to be better medium for more number of shoots induction compared to all other concentrations of IAA + Kn. Maximum number of shoots was observed in the combination of IAA 1.0 mg/1 + BAP 5.0 mg/1. Thus the plant regeneration was established on MS +1.0 mg/1 IAA + 2.0mg/1 BAP in tomato cv pusa ruby from hypocotyls explants, successfully.
