Spinal cord injury (SCI) causes permanent neurological problems that in severe cases may lead to complete palsy. Groups of 6 pound dogs each were subjected to left lateral section of the spinal cord at the lumbar vertebrae (L2) under anesthesia. One group was then implanted with an 50 u.l culture medium of 5x106 bone marrow mesenchymal stem cell scaffold seeded by 0.01mg urinary bladder sub mucosa powder into the sectioned site. Controls were treated with phosphate buffer saline. Dogs were euthanizedat 112 days after section. We monitored improved somatosensory parameters and hind limb performance from the sectioned site by Tarlov and Texas Spinal Cord Injury Scale compared with saline controls (P<0.05). Compared with saline controls, we observed reduced tissue loss from secondary processes, diminished glial scarring tissue, prominent re-myelination of regenerative nerve fibers and improved angiogenesis.
