摘要

In this work, we designed and developed a temperature control system for the polymerase chain reaction (PCR) machine for cell-SELEX-based automated nucleic acid aptamers screening. A 500-mu L PCR reaction chamber was designed for the temperature control system according to the cell-SELEX screening protocol. For the large-volume PCR reaction, a cascade control scheme was adopted for fast and accurate temperature control by combining a fuzzy PID controller with a PI controller. The experimental results showed that the system presented in this paper can implement effective PCR temperature control to meet the design requirements for automation of aptamer screening.
