摘要

L-asparaginase has a pertinent role in the treatment of acute lymphoblastic leukemia by acting as an anti-neoplastic agent for chemotherapy. In the present study production of intra cellular L-asparaginase from Aspergillus fumigates ASPF-6 under submerged fermentation was optimized using Plackett-Burman response surface methodology. The obtained results revealed the two fold increase of asparaginase activity from 17.43U/mL to 36.64 U/mL was due to the change in concentration of carbon source, nitrogen source, Inducer, potassium chloride, incubation period and agitation speed. For substantiating the result further optimization was carried out with multivariate statistical approach using Box-Behnken response surface methodology and an increased activity of 38.63U/mL was observed. Based on the present study fungi Aspergillus fumigatus ASPF-6 isolated from the soil of Kadalundi mangrove forest can be exploited as a potential source for large-scale production of L-asparaginase enzyme based on the needsof industrial application and will able to meet the demand of the global market.
