摘要

This research aims to characterize lipase of Lactococcus garvieae isolated from soil of palm oil mill effluent (POME). Screening of lipolytic bacterial was performed in Rhodamin-B agar plate and olive oil as a substrate. The lipolytic bacteria was characterized by morphology, physiology, and molecular. The lipase activity in various pH and temperature were determined toward p-Nitrophenyl palmitate substrate. The gene fragment of lipase was amplified by PCR method using specific degenerate primer forlipase group IV (HSL). The result of this research showed that one of lipolytic bacteria isolates L49 isolate had highest activity was identified as Lactococcus sp. The lipase from Lactococcus sp. show maximum activity of 9.33 U/mL at pH 7 and temperature 50 °C. Molecular analysis 16S gene of lipolytic bacteria had 85 % homology to Lactococcus garvieae strain FMA401. The fragment lipase gene was 248 bp had 95 % homology with Acinetobacter baumanii strain ab736.
