TESTING OF Escherichia coli IN THE POND WATER WITH MOST PROBABLE NUMBER (MPN) METHOD

Water quality is not only determined by its physical properties, but also content of microorganisms causing disease (pathogens), and the content of chemicals that damage health. Microorganisms as indicators of water quality are coli fecal and coli non-fecal. The research was aimed to test the Escherichia coli to the pond water by MPN method. There are 6 samples. Testing of water quality is conducted by standard methods of APHA (American Public Health Association) to determine the number of coliform bacteria by using MPN table in 3 tubes series includes Presumptive, Confirmed and Completed Tests. The parameters used in this study were Escherichia coli count. The data was analyzed descriptively by seeing MPN table. Then, the data of result were compared with SNI 01-2332.1-2006 standard that still fulfilled the test standard of Escherichia coli is < 3. The results indicated that for several samples analyzed with codes A, B and E are obtained result that still fulfill the standard, while samples with codes C, D and F based total tubes of EC Broth positive continued to IMVIC test, code C is 23 MPN/g, D is 11 MPN/g and F is 1100 MPN/g, does not fulfill the standard or contaminated with Escherichia coli.
