Whole cells of Lactobacillus plantarum A6-1F were used as the catalyst to produce CLA from linoleic acid. To enhance the activity of cell enzymes, permeabilization technology was used. Data showed that freeze-thawing was a suitable and economical permeabilization method for modified L. plantarum, which increased yield by approx. 20%. pH, temp., substrate concn. and cell concn. were the main variables affecting yield of CLA and the reaction conditions of A6-1F were studied. Under optimum reaction conditions (pH 7.0, 15% (w/v) cell concn., 1.5 mg/ml substrate concn. at 37 degrees C), the max. production of CLA (275.7 mug/ml) was obtained in PBS after 2 h reaction. The main CLA isomer in the reaction mixture was c9,t11 CLA which displayed physiological activity.
