摘要

Polymerase chain reaction (PCR) amplification of DNA from formaldehyde-fixed and paraffin-embedded tissue is a relatively simple and sensitive method to investigate large numbers of samples. Existing published methods for sample preparation of DNA from fixed and embedded tissues are successful and are routinely employed for PCR amplifications, These methods allow amplification with high yield of several hundred base pairs of DNA from tissue sections de-paraffinized with xylene and then subjected to protein digestion before PCR, One potentially problematic step with these techniques involves the deparaf-finization of the tissue sections, prior to isolating genomic DNA and subsequent amplification by PCR, De-paraffin izati on of the sections with organic solvents is time-consuming, laborious and requires several centrifu-galions and washes, during which there is the potential for the introduction of contaminants. There is no published technique that avoids these difficulties without impacting the yield.
