A study was conducted to establish the most reliable and sensitive serological and molecular protocols that detect the presence of the most important viruses causing severe problems in the cultivation of artichoke, particularly artichoke latent virus (ArLV), the most widely distributed virus. Different diagnostic methods (one-step RT-PCR and tissue imprint molecular hybridization with DNA probe) were applied to detect ArLV both in vivo and in vitro-cultured 'late' artichoke germplasm. Contemporarily, RT-PCR procedures and ELISA tests were evaluated on a large number of field samples for the diagnosis of artichoke mottled crinkle virus (AMCV), artichoke Italian latent virus (AILV), tomato spotted wilt virus (TSWV), Impatiens necrotic spot virus (INSV) and cucumber mosaic virus (CMV). ArLV was identified in 83% of assayed samples. AMCV was detected in a limited number of plants from 2 fields near Rome while AILV was detected in 3.5% of the total collected samples. TSWV was found in a high percentage in some field in all areas. INSV and CMV were never detected. No differences were observed between one-step RT-PCR and tissue imprint hybridization in revealing the presence of ArLV from leaf samples. ArLV was detected in the same number of plants by the two methods. ELISA test was efficient in detecting the presence of TSWV from leaf tissues. One-step RT-PCR was sensitive and reliable in detecting AMCV, AILV and ArLV in field-collected 'late' artichoke samples.
