In this issue ofCancer Cell, Brunetti and colleagues elucidate the role of mutant NPM1c and its cytoplasmic mislocalization in acute myeloid leukemia. They demonstrate how mutant-specific degradation or relocalization leads to a loss of the stem cell signature characteristic of these leukemias and induces their differentiation.
