Inheritance of wilt resistance in chickpea - a molecular marker analysis.

Mayer et al. developed an allele-specific associated primer (ASAP) marker (CS-27700), which identifies in chickpea germplasm already established locus (H1) susceptible to race 1 of Fusarium wilt pathogen, Fusarium oxysporum f.sp. ciceris. An attempt has been made to validate this marker in a cross between highly susceptible (JG-62) and resistant (WR-315) chickpea genotypes. An attempt was also made to understand the inheritance of wilt resistance coupled with segregation of associated marker. The parental genotypes showed polymorphism for the ASAP marker. Among 100 F5 progenies, 53 showed the presence of CS-27700 marker linked to H1 and in the remaining progenies, the marker was absent. The goodness-of-fit for 1:1 segregation ratio showed single locus segregation of the linked marker, thus supporting the contention that the parents differ at the locus linked to the marker. Among the 100 F5 progenies studied under pot culture, 28 showed early-wilting (took less than 25 days for complete wilt), 43 showed late-wilting (25-55 days), while 29 showed no wilting (no symptoms of wilting on the 55th day), segregating in the ratio of 1:2:1 in the F5 generation, thus indicating that more than one gene is involved in resistance.
