RP [reversed phase]-HPLC and RP-TLC [thin layer chromatography] were used for the analysis of podophyllotoxin in the underground parts of P. hexandrum collected from Koksar, Kullu, Himachal Pradesh, India. HPLC and HPTLC data were also compared to identify an accurate, specific and reproducible method for podophyllotoxin analysis. The analysis of podophyllotoxin in extracts and resins was performed using the external standard method, with pure podophyllotoxin as the standard. The amount of podophyllotoxin in the methanol extract ranged from 4.25 to 5.22% and from 4.15 to 5.12% using HPLC and HPTLC, respectively. Beyond these limits, there were difficulties in extraction and consistency of results. HPLC and HPTLC revealed that the content of podophyllotoxin in the resin reached 4.91-5.57 and 4.90-5.58%, respectively. A good correlation between values was obtained, and a high correlation was evident between methods. The results suggest that HPTLC is a simple and rapid method for the routine analysis of the podophyllotoxin content of a large number of samples.
