摘要

The scope and importance of fluorescence-based detection of DNA in understanding genome architecture is discussed under the following headings: basic technique of fluorescence in situ hybridization (FISH); and analysing genome architecture through FISH, including genome in situ hybridization (identification of genomes in hybrids, determination of ancestry, monitoring of alien chromatin/translocations/intergenomic pairing, chromosome painting and diagnostic cytogenetics, and elucidation of species relationships through cross hybridization of probe), and FISH and physical mapping (repetitive DNA sequences, ribosomal RNA genes, tandem repeats, telomeric sequences, B chromosome specific repeats, centromeric sequences, microsatellites and mapping of low/single copy sequences).
