The effects of the state of cell spreading,on solid surfaces displayed by recombinant Chinese hamster ovary(rCHO)cells genetically modified to produce granulocyte colony-stimulating factor(G-CSF),on growth and G-CSF productivity was examined in a static culture.The change in cell spreading during growth was observed in a plastic tissue culture well having its surface coated with poly-2-hydroxyethyl-methacrylate(pHEMA),a cell attachment inhibitor.Cells were observed to change from well spread to spherical in accord with the concentration of the pHEMA treatment.Spherically growing cells were also induced by adding fibronectin active tetrapeptide Arg-Gly-Asp-Ser(RGDS),a cell attachment factor,into the culture medium.Regardless of the cell spreading or the presence of RGDS,similar growth characteristics were obtained for rCHO cells,while the productivity of G-CSF in the culture increased with the increase of the extent of cell spreading on the surface of the culture well.The maximum G-CSF productivity of the well spread cells in the non-treated culture(control)was 1.5 times higher than that of suspended spherical cells.Cells that spread well on the culture well surface enhanced the production of cellular metabolites such as G-CSF.
