Cotyledon and hypocotyl explants from 5- to 6-day-old sterilized seeds of cv. Snowball were cultured on 4 MS-based media incubated in light at 25degC under 16 h photoperiods. MS medium supplemented with 0.1 mg IAA and 1.0 kinetin/litre was the best, resulting in shoot formation from 100% of cultured explants (both cotyledons and hypocotyls); shoots per explant averaged 6-8 from hypocotyl explants and 8-10 from cotyledon explants. Cotyledons produced shoots in a much lesser time than hypocotyls (10-15vs. 20-25 days, respectively).
