This article reviews the progress in the technology and application of mass spectrometry (MS)-based quantitative proteomics since 2009. The review starts by surveying the established technologies for quantitative proteomics (categorized according to whether or not a chemical derivatization step is involved) and the selected applications for emphasizing the characteristic attributes of a technique. Following a focused discussion on methods in proteomic quantitation that utilize small molecule probes, the review concludes with the author's perspectives of the ever-improving analytical performance for quantitative proteomics platforms. This review is not intended to be comprehensive. Since the publication of the last Analytical Chemistry review on proteomics in 2009, there has been an explosive growth in the literature of quantitative proteomics. A SciFinder search has resulted in more than 1500 journal articles containing the key word or the concept "quantitative proteomics" and more than 250 reviews, appreciating the booming landscape of quantitative proteomics from a variety of standpoints including the informatics advance.
