Posttranslational modification of proteins with ubiquitin (Ub) and ubiquitin-like (Ubl) modifiers, such as SUMO (small ubiquitin-like modifiers), play an essential role in the regulation of numerous cellular pathways, which include proteasomal degradation, nuclear transport, transcription, and DNA repair. Ubl modifiers are typically conjugated by an isopeptide bond, which is formed between the C-terminal carboxyl group of the modifier and the e-amino group of an acceptor lysine (Lys) residue in the substrate protein. Ubiquitin can form poly-ubiquitin chains through all of its seven lysine units. Humans encode four SUMO isoforms, namely SUMOl to SUMO4.
