APP leads to at least some histopathological features of IBM (18, 26). Recent studies by others and us in nonmuscle cells have demonstrated that increased dietary intake of cholesterol can lead to increased Abeta production and conversely that decreased levels of cholesterol can reduce Abeta generation (10, 13, 14, 22, 23). Further implicating cholesterol in APP processing are findings from IBM muscle fibers that accumulated free cholesterol colocalizes with Abeta, hyperphos-phorylated tau, and the intracellular transporter of cholesterol caveolin-1 (16).
